The fine-tuning of cell membrane lipid bilayers accentuates their compositional complexity

Tamir Dingjan et al.

Summary: Cell membranes are now seen as finely tuned molecular systems, with their structural, compositional, and functional complexity often surprising cell and molecular biologists. Models of lipid bilayer structure have evolved considerably, especially with the discovery of restricted diffusion of membrane proteins and lipids within the bilayer. Refinement of current models and describing lipid bilayers as finely tuned molecular assemblies are proposed to better capture their complexity and function.

The noncanonical chronicles: Emerging roles of sphingolipid structural variants

Brenda Wan Shing Lam et al.

Summary: Sphingolipids (SPs) are structurally diverse and abundant in mammalian cells, with many species serving as bioactive mediators of essential cellular processes. Studies have found clinical associations between noncanonical SPs and diseases, suggesting functional significance in SP backbone structure alterations. Further research on noncanonical SPs may provide new insights into pathophysiological processes and potential biomarkers for disease, leading to novel therapeutic strategies.

Structural insights into the regulation of human serine palmitoyltransferase complexes

Yingdi Wang et al.

Summary: The cryo-EM structures of enzyme complexes involved in the rate-limiting step of sphingolipid synthesis reveal mechanisms of substrate recognition and modulation by regulatory subunits. The serine palmitoyltransferase holocomplex consists of catalytic and regulatory components, with regulatory components controlling enzyme activity by modulating the substrate-binding tunnel. These findings provide insights into sphingolipid biogenesis governed by the serine palmitoyltransferase complex.

Ceramide synthases: Reflections on the impact of Dr. Lina M. Obeid

Jiyoon L. Kim et al.

Summary: Sphingolipids are a crucial family of lipids in cell function and survival, and recent research in this field has been led by a closely-knit group of researchers, including the late Dr. Lina Obeid. Dr. Obeid's valuable contributions to sphingolipid research centered on the "many-worlds" view of ceramides and the role of ceramide synthases (CerS) in the sphingolipid metabolic pathway.
