PKCε stimulation of TRPV1 orchestrates carotid body responses to asthmakines

Nicholas G. Jendzjowsky et al.

Summary: The carotid bodies can be stimulated by asthma-associated Th2 cytokines, leading to sensory nerve excitation and asthmatic bronchoconstriction. Inhibiting the PKC ε-TRPV1 signaling pathway may alleviate respiratory distress and airway hyperresponsiveness in asthmatic patients.

CAROTID BODY CHEMORECEPTORS: PHYSIOLOGY, PATHOLOGY, AND IMPLICATIONS FOR HEALTH AND DISEASE

Rodrigo Iturriaga et al.

Summary: This review discusses the role of the carotid body in maintaining homeostasis through reflex ventilatory, cardiovascular, and humoral responses. It also examines the transduction mechanisms, synaptic mechanisms, and integrated reflex responses to CB activation in different physiological, pathological, or environmental challenges. The contribution of enhanced CB chemosensory discharge to autonomic and cardiorespiratory pathophysiology in various diseases is also highlighted.

Oxygen-dependent regulation of ion channels: acute responses, post-translational modification, and response to chronic hypoxia

Summary: Oxygen is crucial for cell survival in aerobic organisms, and lack of oxygen leads to hypoxia. Neural control mechanisms ensure adequate oxygen distribution to cells and tissues. Research on the modulation of oxygen distribution systems and the sensing mechanisms of oxygen levels is of great interest.

Lactate sensing mechanisms in arterial chemoreceptor cells

Hortensia Torres-Torrelo et al.

Summary: Lactate, traditionally seen as a by-product of anaerobic metabolism, is now understood to be a crucial fuel for mitochondrial oxidative phosphorylation and a signaling molecule with medical significance. Changes in blood lactate levels can trigger adaptive responses in normal and pathological conditions. Carotid body cells are shown to sense lactate and hypoxia through separate mechanisms, but activate the same pathway to enhance compensatory respiratory reflexes.

Mitochondrial Succinate Metabolism and Reactive Oxygen Species Are Important but Not Essential for Eliciting Carotid Body and Ventilatory Responses to Hypoxia in the Rat

Agnieszka Swiderska et al.

Summary: The study suggests that alterations in succinate metabolism and mitochondrial ROS generation can modify carotid body nerve activity and whole body ventilatory responses to hypoxia in rats. This indicates that these factors play a role in oxygen sensing in rats, but are not the sole mediators. Antioxidant treatment of the carotid body with mitochondrial selective antioxidants may offer a new approach for treating cardiovascular-respiratory disorders related to carotid body dysfunction.

Summary: This protocol allows for monitoring acute changes in ROS production in sensory cells from mouse carotid body slices using redox-sensitive green fluorescent protein probes and microfluorimetry, providing sensitive and reproducible quantification of ROS during acute hypoxia in different subcellular compartments of CB glomus cells.
