Chemoprotective Antimalarial Activity of P218 against Plasmodium falciparum: A Randomized, Placebo-Controlled Volunteer Infection Study

Mohamed Farouk Chughlay et al.

Summary: P218 demonstrated excellent chemoprotective efficacy against P. falciparum, effectively inhibiting the parasite. Clinical trials showed that P218 had good safety and tolerability in participants, with mild, transient, and self-limiting adverse events. Furthermore, P218's pharmacokinetics were similar in participants with and without induced infection.

Transgenic pyrimethamine-resistant plasmodium falciparum reveals transmission-blocking potency of P218, a novel antifolate candidate drug

Navaporn Posayapisit et al.

Summary: Antimalarial drugs targeting multiple parasite stages, especially the transmissible stages, are essential for malaria elimination. Developing new antifolate compounds to overcome antifolate resistance, including in mosquito development stages, is critical. The use of genome editing to create a pyrimethamine-resistant transgenic strain of Plasmodium falciparum provides a robust system for evaluating novel antifolate compounds.
