Three-dimensional superresolution fluorescence microscopy maps the variable molecular architecture of the nuclear pore complex

Vilma Jimenez Sabinina et al.

Summary: NPCs are large macromolecular machines that regulate traffic between the nucleus and the cytoplasm, consisting of about 1000 proteins and having a mass of over 100 MDa in vertebrates. While a static model of the central scaffold of the NPC has been assembled, many structural components remain unknown due to the NPC's size and flexibility. This study combined 3D super-resolution microscopy with computational classification to create a detailed 3D structural map of the NPC in single human cells, providing insight into the different conformations of the nuclear ring and the nuclear basket.
