Prognosis and personalized treatment prediction in TP53-mutant hepatocellular carcinoma: an in silico strategy towards precision oncology

Chen Yang et al.

Summary: This study retrospectively analyzed 1135 HCC patients and developed a random forest-based prediction model to estimate TP53 mutational status, as well as a robust poor prognosis-associated signature which showed superior ability to predict survival in TP53-mutant patients. In silico screening revealed three targets and two agents that might have potential therapeutic implications in high-PPS patients, presenting a comprehensive view of potential treatment strategy.

Identification of the pyroptosis-related prognostic gene signature and the associated regulation axis in lung adenocarcinoma

Wanli Lin et al.

Summary: A prognostic PRG signature containing five genes (NLRP7, NLRP1, NLRP2, NOD1, and CASP6) was identified for LUAD patients through comprehensive bioinformatics analysis. Additionally, a lncRNA KCNQ1OT1/miR-335-5p/NLRP1/NLRP7 regulatory axis was discovered, which may play a significant role in the progression of LUAD. Further research is needed to validate these findings.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.

Lighting a Fire: Can We Harness Pyroptosis to Ignite Antitumor Immunity?

Zhibin Zhang et al.

Summary: The research highlights the role of gasdermin-mediated inflammatory death in promoting antitumor immunity and converting immune cell killing to inflammatory pyroptosis. This may enhance the effectiveness of immunotherapy but also increase therapy-related toxicity, requiring further study to understand its regulation and potential therapeutic applications.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

A novel defined pyroptosis-related gene signature for predicting the prognosis of ovarian cancer

Ying Ye et al.

Summary: This study identified pyroptosis-related genes in ovarian cancer (OC) that may play important roles in tumor immunity and can be used to predict the prognosis of OCs. By dividing OC cases into subtypes based on the expression of these genes, different risk groups were identified with significant differences in survival possibilities. Additionally, immune-related genes were found to be enriched in high-risk groups, indicating a decreased immune status in these patients.

The Pyroptosis-Related Signature Predicts Prognosis and Indicates Immune Microenvironment Infiltration in Gastric Cancer

Wei Shao et al.

Summary: This study classified gastric cancer patients based on pyroptosis-related regulators and developed a prognostic model called PS-score, which can predict patients' prognosis and overall survival time. Patients with a low PS-score have more inflammatory cell infiltration and better response to PD1/PD-L1 blockade immunotherapy.

NLRP3 inflammasome-mediated cytokine production and pyroptosis cell death in breast cancer

Sara Socorro Faria et al.

Summary: Chronic inflammation plays a crucial role in breast cancer pathobiology, affecting tumor progression, metastasis, and therapeutic outcomes. Inflammasomes are protein complexes that respond to damage- and pathogen-associated molecular patterns, triggering the release of pro-inflammatory cytokines. Dysregulation of inflammasome activation can lead to inflammatory diseases, neurodegeneration, and cancer.

Summary: This study identified the tumor suppression role of DRD2 in BrCa through downregulation due to promoter methylation, correlation with longer survival in HER2-positive patients, promotion of drug sensitivity, inhibition of tumorigenesis, induction of apoptosis and necroptosis, interaction with various genes to restrict signaling pathway activation, modulation of the microenvironment, and providing predictive and therapeutic targets for BrCa.

Cisplatin Induces Pyroptosis via Activation of MEG3/NLRP3/caspase-1/GSDMD Pathway in Triple-Negative Breast Cancer

Honglin Yan et al.

Summary: The study revealed that cisplatin induces pyroptosis in triple-negative breast cancer cells through activation of the MEG3/NLRP3/caspase-1/GSDMD pathway, leading to its anti-tumor effects. This finding may contribute to the development of new therapeutic strategies for TNBC.
