Phosphoric Acid Mediated Light-Induced Minisci C-H Alkylation of N-Heteroarenes

Summary: In this study, an environmentally-friendly light-induced Minisci alkylation of N-heteroarenes using diphenyl phosphate as catalyst under metal- and photocatalyst-free conditions was reported. The use of radical precursor redox-active esters (RAEs) as alkylating reagents for the functionalization of N-heteroarene derivatives was demonstrated, with a focus on pyridine, quinoline, and isoquinoline. Mechanistic studies indicated that diphenyl phosphate played a key role through hydrogen bonding in the catalytic cycle.

Summary: The addition of tertiary carbon radicals from N-(acyloxy)phthalimide esters to cyclic alpha, beta-unsaturated ketones and lactones is greatly enhanced by substoichiometric amounts of Ln(OTf)(3). The reaction, which does not require a photosensitizer, is suggested to proceed through excitation of a ternary electron donor-acceptor complex between the NHPI ester, Hantzsch ester, and Ln(OTf)(3).

Hydrogen Atom Transfer-Driven Enantioselective Minisci Reaction of Amides

Summary: Minisci-type reactions are powerful methods for building complexity around basic heteroarenes. The disclosed method enables coupling of linear amides and heteroarenes with control of enantioselectivity at the newly formed stereocenter and site selectivity on both the heteroarene and the amide. This is achieved using a chiral phosphoric acid catalyst in conjunction with diacetyl as a combined hydrogen atom transfer reagent and oxidant, without the need for an extraneous photocatalyst.
