Mechanical stretch induces osteogenesis through the alternative activation of macrophages

Summary: This study reveals that macrophages stimulated with mechanical stretch under specific conditions can efficiently induce osteogenic differentiation of MSCs through a synergy mechanism between M2 polarization and YAP/BMP2 axis. Additionally, the mechanical stretch activates macrophages to produce anti-inflammatory cytokines, while also inducing YAP activation and downstream BMP2 expression to facilitate MSCs osteogenesis. These findings advance understanding of the complex influence among mechanical strain, macrophage inflammatory response, and MSC osteogenic differentiation, providing insights into the control system from mechanical signals to chemical response during bone repair and regeneration.
