Recent Advances in Hyperthermia Therapy-Based Synergistic Immunotherapy

Mengyu Chang et al.

Summary: Hyperthermia therapy has emerged as a strategy against malignant tumors, particularly when combined with nanomaterial-based photothermal therapy or magnetic hyperthermia. While these treatments alone may not completely cure cancer, when combined with cancer immunotherapy, they have shown potential for enhanced therapeutic outcomes against both primary tumors and metastatic lesions, as well as preventing cancer recurrence and prolonging survival period.

Activatable Polymer Nanoenzymes for Photodynamic Immunometabolic Cancer Therapy

Ziling Zeng et al.

Summary: This study introduces an organic polymer nanoenzyme (SPNK) with near-infrared (NIR) photoactivatable immunotherapeutic effects for photodynamic immunometabolic therapy. By generating singlet oxygen (O-1(2)) upon NIR photoirradiation, SPNK induces immunogenic cell death of cancer and degrades immunosuppressive kynurenine (Kyn), ultimately promoting effector T cell proliferation and infiltration, enhancing systemic antitumor T cell immunity, and inhibiting both primary and distant tumors in living mice.

Yan Zhang et al.

Summary: The oxygen-vacancy molybdenum trioxide nanodots (MoO3-x NDs) possess triple-therapy synergistic efficiency based on the single near-infrared irradiation (808 nm) regulated combination of photodynamic, photothermal, and peroxidase-like enzymatic activities, showing excellent antibacterial efficiency against drug-resistant bacteria.

A Polymer Multicellular Nanoengager for Synergistic NIR-II Photothermal Immunotherapy

Cheng Xu et al.

Summary: SPNE is a polymer multicellular nanoengager that combines an NIR-II absorbing polymer core with fused membranes from immunologically engineered tumor cells and DCs for synergistic second-NIR-II photothermal immunotherapy. It enables effective interactions among tumor cells, DCs, and T cells by high accumulation in lymph nodes and tumors, leading to enhanced T cell activation.

Engineering Oxaliplatin Prodrug Nanoparticles for Second Near-Infrared Fluorescence Imaging-Guided Immunotherapy of Colorectal Cancer

Qiurong Zhu et al.

Summary: A versatile prodrug nanoparticle for second near-infrared (NIR-II) fluorescence imaging-guided combinatory immunotherapy of colorectal cancer has been reported. The nanoparticles show high intratumoral accumulation and trigger immunogenic cell death of the tumor cells, elicit antitumor immune response in a spatiotemporally controllable manner. The combination of the nanoparticle-based photothermal therapy/chemotherapy with programmed death ligand 1 blockade significantly promotes intratumoral infiltration of cytotoxic T lymphocytes and eradicates CRC tumors.

Davide Porrelli et al.

Summary: The study aimed to enhance the bioactivity of electrospun PCL membranes by coating them with bioactive lactose-modified chitosan (CTL), which can be used for the synthesis and stabilization of silver nanoparticles to provide antibacterial properties. The CTL coating improved MG63 osteoblast adhesion and proliferation, while discouraging biofilm formation by Pseudomonas aeruginosa and Staphylococcus aureus.

Diclofenac Resensitizes Methicillin-Resistant Staphylococcus aureus to β-Lactams and Prevents Implant Infections

Shutao Zhang et al.

Summary: The study shows that high-dose diclofenac inhibits the growth of MRSA without inducing drug-resistant mutations easily, while low-dose diclofenac can resensitize bacteria to beta-lactams and inhibit biofilm formation. Transcriptomic and proteomic analyses indicate that diclofenac reduces the expression of genes and proteins associated with antibiotic resistance and biofilm formation. Murine implant infection models suggest that diclofenac combined with beta-lactams can substantially alleviate MRSA infections in vivo, offering promising applications for preventing perioperative infections.

A Biomimetic Aggregation-Induced Emission Photosensitizer with Antigen-Presenting and Hitchhiking Function for Lipid Droplet Targeted Photodynamic Immunotherapy

Xiuli Xu et al.

Summary: The biomimetic aggregation-induced emission (AIE) photosensitizers with antigen-presenting and hitchhiking abilities (DC@AIEdots) were developed by coating dendritic cell (DC) membranes on the nanoaggregates of the AIEgens. This technique can enhance tumor delivery efficiency by about 1.6 times and stimulate in vivo proliferation and activation of T cells. Additionally, a potential application of therapeutic agents targeting lipid droplets for immunotherapy is indicated, providing a new hitchhiking approach for drug delivery.

Restoring the Immunity in the Tumor Microenvironment: Insights into Immunogenic Cell Death in Onco-Therapies

Angela-Patricia Hernandez et al.

Summary: The concept of cancer as a single cell mass that replicates unlimitedly in isolation has been dissolved with the inclusion of immune evasion as a hallmark. Tumor microenvironment (TME) plays a crucial role in drug resistance, impacting the success of treatment. Immunogenic cell death (ICD) elicited by cancer therapy reshapes the tumor immune microenvironment, initiating a long-term adaptative immune response.

Activating an adaptive immune response from a hydrogel scaffold imparts regenerative wound healing

Donald R. Griffin et al.

Summary: The study aimed to slow MAP scaffold degradation by changing the chirality of peptides, but unexpectedly, D-peptide crosslinked MAP scaffolds led to accelerated degradation in vivo, promoting tissue regeneration and enhancing wound healing. D-MAP triggered an immune response against D-chiral peptides and intact adaptive immune system was crucial for skin regeneration induced by the hydrogel.
