Production, Processing, and Characterization of synthetic AAV Gene Therapy Vectors

Jihad El Andari et al.

Summary: Over the past two decades, gene therapy vectors based on wild-type AAV have proven safe and effective in clinical trials, while engineered synthetic AAV capsids show promising potential. There is a growing demand for new processes to accommodate the expanding range of AAV capsid variants.

Development of an optimized and scalable method for isolation of umbilical cord blood-derived small extracellular vesicles for future clinical use

Renato M. S. Cardoso et al.

Summary: The article presents an optimized and scalable process for isolating extracellular vesicles (EV) smaller than 200 nm, secreted by umbilical cord blood mononuclear cells (UCB-MNC), providing a method foundation for further research in regenerative medicine. The use of ultrafiltration and size exclusion chromatography (UF/SEC) showed superior benefits compared to traditional ultracentrifugation (UC), allowing for increased vesicle yield and potential for mass production in a clinical setting.

Critical considerations for the development of potency tests for therapeutic applications of mesenchymal stromal cell-derived small extracellular vesicles

Mario Gimona et al.

Summary: Mesenchymal stromal/stem cells (MSCs) have been approved for treating certain diseases through paracrine secretion of small extracellular vesicles (sEVs). Despite the potential advantages of sEV preparations over cellular MSC products, there are challenges in developing MSC-sEV drug products, including establishing quality control metrics and developing effective potency assays.

Summary: Extracellular vesicles (EVs) are complex nanoparticles that have shown potential as natural platforms for enhanced therapeutic delivery across biological barriers. However, there is currently no standard reporting framework established for therapeutic loading of EVs for drug delivery applications.

Small extracellular vesicles obtained from hypoxic mesenchymal stromal cells have unique characteristics that promote cerebral angiogenesis, brain remodeling and neurological recovery after focal cerebral ischemia in mice

Jonas Gregorius et al.

Summary: Obtained from the right cell-type, mesenchymal stromal cell (MSC)-derived small extracellular vesicles (sEVs) promote stroke recovery by enhancing microvascular remodeling. In vitro, sEVs from hypoxic MSCs, but not 'normoxic' MSCs, dose-dependently promoted endothelial proliferation, migration, and tube formation, as well as increased post-ischemic endothelial survival.

Protein adsoprotein adsorption on core-shell particles: Comparison of CaptoTM Core 400 and 700 resins

Xue Mi et al.

Summary: The structural and functional characteristics of two core-shell resins, Capto Core 400 and 700, were compared for the purification of bioparticles. While Capto Core 400 has higher BSA binding capacity, Capto Core 700 shows greater capacity for larger proteins like Tg. Mass transfer in both resins is influenced by diffusional resistances, with different effective pore diffusivities for small and large proteins.

Eribulin and Paclitaxel Differentially Alter Extracellular Vesicles and Their Cargo from Triple-Negative Breast Cancer Cells

Petra J. Pederson et al.

Summary: The study investigated the impact of microtubule-targeting drugs eribulin and paclitaxel on the release and cargo of extracellular vesicles from breast cancer cells. While both drugs did not alter the concentration of vesicles released, eribulin reduced the levels of ILK, a key cargo promoting cancer progression. This differential effect highlights the potential clinical significance of drug-induced changes in extracellular vesicles and their cargo. Overall, the findings suggest that eribulin can specifically reduce certain cargo in extracellular vesicles, potentially contributing to its efficacy in reversing epithelial-to-mesenchymal transition in breast cancer.
