Dual-targeting Approach on Histamine H3 and Sigma-1 Receptor Ligands as Promising Pharmacological Tools in the Treatment of CNS-linked Disorders

Katarzyna Szczepanska et al.

Summary: The market approval of Pitolisant has increased interest in clinical applications for novel multifunctional histamine H3 receptor antagonists. Various combinations of H3R pharmacophores with elements of other receptors, transporters, or enzymes have been synthesized. Recent studies suggest that some H3 receptor antagonists have affinity for sigma-1 receptor binding sites, which may impact their overall efficacy. The combination of histamine H3 and sigma-1 receptors could be a potential target for therapeutic developments in treating various diseases.

Sigma-1 receptor: A drug target for the modulation of neuroimmune and neuroglial interactions during chronic pain

Summary: This article discusses the modulatory effects of sigma-1 receptors on communication between the immune and nervous systems during inflammation, as well as their influence on peripheral and central neuroinflammation. Experimental models show that sigma-1 receptor inhibition can prevent neuroinflammation and enhance immune-driven opioid analgesia, potentially making sigma-1 antagonists a new class of analgesics for various painful disorders.

Structural modifications in the distal, regulatory region of histamine H3 receptor antagonists leading to the identification of a potent anti-obesity agent

Katarzyna Szczepanska et al.

Summary: A study on 4-pyridylpiperazine derivatives as histamine H-3 receptor ligands showed that the introduction of electron-withdrawing moieties into the distal aromatic ring significantly affected their affinity toward H3R. The 4-pyridylpiperazine moiety was found to be crucial for suitable interaction with the human histamine H-3 receptor, with even slight structural modifications leading to reduced affinity for H3R. Additionally, molecular modeling techniques were used to demonstrate the protein-ligand interactions responsible for the high affinity observed.

Summary: The novel fluorescent H3R ligand UR-NR266 has been synthesized and characterized for its exceptional selectivity and pharmacological properties at the sub-nanomolar level. It shows fast association and dissociation kinetics at the H3R, making it a versatile pharmacological tool for analyzing future H3R ligands. UR-NR266 also exhibits low nonspecific binding and has been identified as a selective compound in off-target screening at 14 receptors.
