Comprehensive analysis of O-glycosylation of amyloid precursor protein (APP) using targeted and multi-fragmentation MS strategy

Jingjing Shi et al.

Summary: This study established a comprehensive O-glycopeptide mass spectrometry analysis strategy for APP O-glycopeptide identification, successfully identifying 14 O-glycosites on APP695 expressed in HEK-293 T cells. The research found at least four O-glycans present at specific residues on APP, providing valuable insights into the O-glycosylation patterns of APP.

In-depth Site-specific Analysis of N-glycoproteome in Human Cerebrospinal Fluid and Glycosylation Landscape Changes in Alzheimer's Disease

Summary: A novel N-glycoproteomic approach was developed in this study and successfully applied to analyze human cerebrospinal fluid samples, identifying a large number of intact N-glycopeptides and providing a molecular basis for understanding the structure-function relationships and disease mechanisms related to glycoproteins in cerebrospinal fluid. Additionally, a comparative in-depth N-glycoproteomic analysis of cerebrospinal fluid samples from healthy controls and Alzheimer's disease patients revealed altered glycosylation patterns for several N-glycoproteins, suggesting potential targets for further studies on glycosylation-based Alzheimer's disease research.
