Advances in Activity-Based Sensing Probes for Isoform-Selective Imaging of Enzymatic Activity

Summary: Activity-based sensing (ABS) has emerged as a powerful approach for monitoring small-molecule and enzyme activities within living systems, with a recent focus on increasing selectivity to monitor a single enzyme or isoform. The highest degree of selectivity is required for differentiating between isoforms, where the targets display significant structural similarities as a result of gene duplication or alternative splicing. This Minireview highlights key examples of small-molecule isoform-selective probes, focusing on the relevance of isoform differentiation, design strategies to achieve selectivity, and applications in basic biology or in the clinic.

Point-of-care semen analysis of patients with infertility via smartphone and colorimetric paper-based diagnostic device

Scaling up COVID-19 rapid antigen tests: promises and challenges

Summary: WHO recommends antigen-detection rapid diagnostic tests (Ag-RDTs) with at least 80% sensitivity and 97% specificity for patients with symptoms consistent with COVID-19. Ag-RDTs may show high false negative rates after the acute phase, suggesting the need for a combination of molecular and serological tests. In low prevalence settings and situations where infection needs to be ruled out, Ag-RDTs with high negative predictive values can be used confidently, but high false positive rates in low prevalence areas require further confirmation.

Chemiluminescence Detection of Hydrogen Sulfide Release by β-Lactamase-Catalyzed β-Lactam Biodegradation: Unprecedented Pathway for Monitoring β-Lactam Antibiotic Bacterial Resistance

Summary: The discovery of a new biodegradation metabolite (H2S) generated through beta-lactamase-catalyzed hydrolysis of beta-lactam antibiotics has led to the development of a distinct molecular technique for monitoring bacterial antibiotic resistance. This technique, based on a chemiluminescence probe designed for the detection of hydrogen sulfide released during biodegradation of beta-lactam antibiotics, has been successfully demonstrated for five different antibiotics and eight resistant bacterial strains. The findings suggest that hydrogen sulfide may be considered an emerging beta-lactam metabolite for detecting bacterial resistance.

Summary: This study systematically compared the diagnostic performance of different clinical specimen collection methods for SARS-CoV-2 infection. Pooled nasal and throat swabs showed the highest sensitivity and positive predictive value, while saliva and nasal swabs also demonstrated good diagnostic performance. Throat swabs had a significantly lower sensitivity and positive predictive value and were not recommended for diagnosis. Self-collection for pooled nasal and throat swabs and nasal swabs did not affect diagnostic accuracy.

FLASH can distinguish live from dead cells and has the potential to enhance TB diagnostics and drug resistance monitoring in resource-limited settings.
