Abundance of P-Glycoprotein and Other Drug Transporters at the Human Blood-Brain Barrier in Alzheimer's Disease: A Quantitative Targeted Proteomic Study

Summary: The study found that the abundance of P-gp in the BBB of the hippocampus was decreased in both subjects with AD and age-matched controls, but it was not related to AD. Additionally, the abundance of P-gp in gray matter BBB of the hippocampus and parietal lobe was not different between AD and controls. The abundance of all drug transporters at the gray matter BBB decreased with age, likely due to age-dependent decrease in brain microvessel density.

Impact of CNS Diseases on Drug Delivery to Brain Extracellular and Intracellular Target Sites in Human: A WHAT-IF Simulation Study

Summary: This study investigated the impact of altered cerebral blood flow, tight junction paracellular pore radius, brain extracellular fluid volume, and pH values on drug transport and concentrations in the central nervous system. The results showed drug-dependent effects of pathophysiological changes on blood-brain barrier transport and drug concentrations within the brain extracellular and intracellular fluid. Changes in pore radius, pH values, and blood flow affected the rate and extent of drug transport, highlighting the importance of considering these factors in drug delivery to the brain.

A physiologically-based pharmacokinetic model to describe antisense oligonucleotide distribution after intrathecal administration

Summary: This study focuses on the potential of antisense oligonucleotides (ASOs) in treating neurodegenerative and neuromuscular disorders, particularly through intrathecal (IT) administration to bypass the blood-brain barrier. It was found that maximum exposure of CNS tissues with IT-administered ASOs can be achieved within two days, and clearance is mainly controlled by the rate of transfer from CNS tissues back to CSF.
