To mimic long-term exposure for toxicity testing, lung epithelial cells must be maintained under ALI conditions and co-cultured with macrophages. Calu-3 cells maintain monolayer structure and tight junctions, making them ideal for co-culturing with macrophages for increased sensitivity in responses to aerosol exposure.
