Summary: This review discusses modern methodological solutions for ensuring high sensitivity of immunochromatographic assay (ICA) methods based on the use of nanoparticle markers and colorimetric detection, focusing on lowering the detection limit amplification of the detectable signal generated by a single nanoparticle. Different approaches are examined for their advantages and limitations, showing that the use of these methods can decrease the detection limit of ICA by 1-3 orders of magnitude, making it competitive with instrumental methods of analysis.

In this study, an ultrasensitive chip-based antigen test with single protein biomarker sensitivity was introduced for differentiated detection of SARS-CoV-2 and influenza A antigens in nasopharyngeal samples. The assay demonstrated a spectrally multiplexed detection of both targets with single-antigen sensitivity at a concentration of 30 ng/mL.
