The role of extracellular vesicles in skeletal muscle and systematic adaptation to exercise

Ivan J. Vechetti et al.

Summary: Regular exercise plays a crucial role in reducing the risk of certain diseases. Skeletal muscle-specific cytokines and extracellular vesicles released during exercise may contribute to the positive effects on metabolism. Research on the potential role of EVs in muscle and systemic adaptation to exercise has generated interest in various fields.

Extracellular Vesicles Derived from Neural Progenitor Cells--a Preclinical Evaluation for Stroke Treatment in Mice

X. Zheng et al.

Summary: Stem cells such as mesenchymal stem cells (MSCs) enhance neurological recovery in preclinical stroke models by secreting extracellular vesicles (EVs), with neural progenitor cells (NPCs)-derived EVs showing therapeutic potential in reducing cell injury and promoting neuroregeneration in mice post-stroke. NPC-EVs delivery leads to increased blood concentrations of B and T lymphocytes without affecting cerebral cell counts, suggesting a potential contribution to reverting post-stroke immunosuppression. Furthermore, biodistribution analysis reveals the majority of NPC-EVs to be found in extracranial organs like the liver and lung after systemic administration.

Microglial vesicles improve post-stroke recovery by preventing immune cell senescence and favoring oligodendrogenesis

Stefano Raffaele et al.

Summary: The activation of microglia and monocyte-derived macrophages affects the regenerative process of oligodendrocyte precursor cells (OPCs), promoting functional recovery after stroke. Regenerative microglia-derived extracellular vesicles (EVs) can restore protective immune cell functions, limit senescence, and enhance OPC maturation, potentially leading to improved neurological functionality in post-stroke phase. Tumor necrosis factor carried by EVs mediates pro-differentiating effects on OPCs, suggesting implications for regenerative therapies.

Neuronal extracellular vesicle derived miR-98 prevents salvageable neurons from microglial phagocytosis in acute ischemic stroke

Jin Yang et al.

Summary: The study demonstrates the protective role of miR-98 in ischemic stroke by inhibiting microglial phagocytosis to reduce neuronal death. Neurons transfer miR-98 to microglia via EVs secretion to prevent phagocytosis of stressed-but-viable neurons.EVs derived miR-98 act as an intercellular signal mediating neurons and microglia communication during brain remodeling after ischemic stroke.

Extracellular vesicles for tissue repair and regeneration: Evidence, challenges and opportunities

Anika Nagelkerke et al.

Summary: Extracellular vesicles (EVs) are biological nanoparticles secreted by cells that act as delivery vehicles for molecular messages, showing potential as therapeutic mediators for various diseases. EVs have regenerative properties and can mediate signaling cascades in target cells to achieve functional outcomes in tissue repair and regeneration. Their interactions and biodistribution in vivo, especially when combined with biomaterials, influence their regenerative effects, with progress being made towards successful implementation of EV regenerative therapies in a clinical setting.

Native and Bioengineered Exosomes for Ischemic Stroke Therapy

Haroon Khan et al.

Summary: Exosomes are natural cells-derived vesicles that are considered as a leading option for treating various diseases, including cerebral ischemia. Despite the potential for ischemic stroke therapy, native exosomes face challenges due to undesirable features. Bioengineering exosomes may enhance their effectiveness in stroke treatment.

Biophotonics for diagnostic detection of extracellular vesicles

Alice Gualerzi et al.

Summary: Extracellular Vesicles (EVs) are versatile carriers for biomarkers associated with multiple human disorders, and the application of biophotonics has led to the development of innovative platforms with excellent sensitivity for their diagnostic detection. These bio-photonic approaches have demonstrated great potential for clinical translation by discriminating between nanovesicles and contaminants, achieving higher resolution than traditional procedures, and showing moderate to excellent diagnostic accuracy in clinical samples.

Small extracellular vesicles obtained from hypoxic mesenchymal stromal cells have unique characteristics that promote cerebral angiogenesis, brain remodeling and neurological recovery after focal cerebral ischemia in mice

Jonas Gregorius et al.

Summary: Obtained from the right cell-type, mesenchymal stromal cell (MSC)-derived small extracellular vesicles (sEVs) promote stroke recovery by enhancing microvascular remodeling. In vitro, sEVs from hypoxic MSCs, but not 'normoxic' MSCs, dose-dependently promoted endothelial proliferation, migration, and tube formation, as well as increased post-ischemic endothelial survival.
