The Role of the Cytoskeleton in Regulating the Natural Killer Cell Immune Response in Health and Disease: From Signaling Dynamics to Function

Summary: NK cells, as innate lymphoid cells, play essential roles in eliminating virally infected and malignant cells, with their immune response regulated by a balance of activating and inhibitory signals from surface receptors. The cytoskeleton is crucial for facilitating most NK cell effector functions, however, the exact mechanisms of how specific cytoskeletal architectures at the immunological synapse promote these functions and impact downstream signaling pathways remain unclear. Pioneering studies using advanced imaging techniques have begun to uncover the complexity of cytoskeletal organization dictating NK cell activation threshold, highlighting the significant role of cytoskeleton organization in NK cell function in health and disease.
