Summary: Cryptococcus species, particularly Cryptococcus neoformans and Cryptococcus gattii, have a significant impact on global human health, with almost a quarter of a million cases and 181,000 deaths from cryptococcal meningitis annually. Despite limited treatment options and high mortality rates, only three major classes of drugs are approved for clinical use. Challenges in developing new therapies include host toxicity, pathogen resistance, and the limited efficacy of existing antifungal agents. Research efforts are focused on understanding resistance mechanisms, exploring novel therapeutic strategies, and accelerating drug discovery through genomics technologies and high-throughput screening.

Increased mortality associated with uncontrolled diabetes mellitus in patients with pulmonary cryptococcosis: a single US cohort study

Summary: Poorly controlled diabetes is associated with increased mortality in pulmonary cryptococcosis patients, with those having uncontrolled diabetes showing significantly higher odds of death at 10 weeks and 1 year compared to controlled or no diabetes individuals. This suggests that interventions to improve glucose control may be important in improving outcomes for patients with pulmonary cryptococcosis.
