Intestinal differentiation involves cleavage of histone H3 N-terminal tails by multiple proteases

Summary: The study found extensive cleavage of histone H3 N-terminal tails in differentiated cells of mouse intestinal epithelium, demonstrating that intestinal H3 clipping is the result of multiple proteolytic activities including Trypsins and Cathepsin L. These proteolytic activities are differentially affected by the PTM pattern of histone H3 tails.

IκBα targeting promotes oxidative stress-dependent cell death

Summary: In lung cancer, an inverse correlation between NFKBIA gene responsible for I kappa B alpha and oxidative metabolism, as well as ROS production, was identified. Additionally, a combination of new I kappa B alpha targeting compounds with cisplatin treatment was found to increase ROS levels beyond a tolerable threshold, leading to oxytosis-induced cell death. Through targeting I kappa B alpha/p65 interactions, the study proposed a novel approach to enhance p65-mediated apoptotic responses to cisplatin by disrupting mitochondrial regulation and ROS production.

Oncohistones are often dominant-negative and more prevalent in tumors affecting children and adolescents. Further studies are needed to understand the mechanisms of action of oncohistones and their potential application in cancer classification and treatment.
