Summary: In this study, the polyphenolic profile of Amontillado sherry was analyzed using high-speed counter-current chromatography (HSCCC) and HPLC-DAD-MS, leading to the identification of 42 compounds, with 11 of them being newly discovered in Sherry wines. Additionally, the antioxidant capacity of the obtained fractions was determined, showing higher values in fractions containing specific compounds like gallic acid, protocatechuic acid, and tyrosol. This is the first known use of HSCCC for characterizing the phenolic composition of Sherry wines.
