Effects of hydrogen sulfide on mitochondrial function and cellular bioenergetics

Bindu D. Paul et al.

Summary: Hydrogen sulfide (H2S) was once thought to be only toxic, but is now known to also act as an endogenous signaling molecule. The effects of H2S on cellular physiology are dose-dependent, with lower concentrations being beneficial and higher concentrations being cytotoxic. In particular, H2S has contrasting effects on mitochondrial function, inhibiting the electron transport chain at higher concentrations and stimulating bioenergetics at lower concentrations.

Mutant Huntingtin stalls ribosomes and represses protein synthesis in a cellular model of Huntington disease

Mehdi Eshraghi et al.

Summary: The study reveals that mHtt promotes ribosome stalling and inhibits protein synthesis in HD cells, with Fmrp being upregulated but not affecting protein synthesis or ribosome stalling. There are interactions between ribosomal proteins, translating ribosomes, and mHtt, leading to a widespread shift in ribosome occupancy and unique single-codon pauses on selected mRNA targets in HD cells. This mechanistic defect of ribosomal translocation during translation elongation can be exploited for HD therapeutics.

Maintenance of organellar protein homeostasis by ER-associated degradation and related mechanisms

Marius K. Lemberg et al.

Summary: Protein homeostasis mechanisms are crucial for meeting cellular needs and dealing with stress, with the ERAD pathway being the best understood organelle protein quality control system. The rhomboid superfamily, including derlins and intramembrane serine proteases, plays a key role in membrane protein quality control and protein dislocation processes, indicating conserved principles across organelles and evolution.

Redox imbalance links COVID-19 and myalgic encephalomyelitis/chronic fatigue syndrome

Bindu D. Paul et al.

Summary: The review summarizes evidence of biological abnormalities in patients with acute COVID-19 and ME/CFS, including redox imbalance, systemic inflammation, neuroinflammation, impaired ATP generation, and general hypometabolism. Although these phenomena have not been extensively studied in individuals with long COVID-19, understanding these abnormalities may lead to the development of novel therapeutics.

DNA Mismatch Repair and its Role in Huntington's Disease

Ravi R. Iyer et al.

Summary: DNA mismatch repair (MMR) is a highly conserved pathway that corrects DNA replication errors, limits chromosomal rearrangements, and responds to various types of DNA damage. Despite its genome stabilizing function, MMR is also involved in generating mutations, such as somatic triplet repeat expansion in neurodegenerative disorders like Huntington's disease. Understanding the mutagenic role of MMR in CAG repeat expansion may provide insights for potential therapeutic interventions in HD.
