Pterostilbene leads to DNMT3B-mediated DNA methylation and silencing of OCT1-targeted oncogenes in breast cancer cells

Summary: The study investigated the dynamics between oncogenic TF OCT1 binding and de novo DNA methyltransferase DNMT3B binding in PTS-treated invasive breast cancer cells. It was found that PTS led to decreased OCT1 binding and increased DNMT3B binding in gene regulatory regions associated with oncogenic functions, resulting in gene methylation and silencing. Our findings provide insights into the mechanistic players and gene targets that contribute to the anticancer action of stilbenoid polyphenols.

Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Summary: Enhancers play a crucial role in gene regulation by increasing the likelihood of gene transcription, and are marked by histone modifications H3K4me1 and H3K27ac. DNA methylation on enhancers has traditionally been associated with repression. This study combined histone marks and DNA methylation to show that H3K4me1-marked enhancers have higher levels of methylation compared to H3K27ac-marked enhancers during mouse embryonic stem cell differentiation, indicating cellular heterogeneity in enhancer states. The enhancers identified in this study are involved in stem cell differentiation and show variation in gene expression levels, highlighting the functional relevance of enhancer-based heterogeneity in cellular dynamics.
