The C-degron pathway eliminates mislocalized proteins and products of deubiquitinating enzymes

Chi-Wei Yeh et al.

Summary: The study elucidates the function of C-degron pathways in removing mislocalized cellular proteins and cleavage products of deubiquitinating enzymes. It also shows that normal proteins targeted by C-degron pathways experience a shortfall, highlighting proteolysis-based immunity as a constraint for protein evolution/selection. The cooperation between C-degron and N-degron pathways in protein removal is also emphasized.

Irreversible incorporation of L-dopa into the C-terminus of α-tubulin inhibits binding of molecular motor KIF5B to microtubules and alters mitochondrial traffic along the axon

Agustina Zorgniotti et al.

Summary: The study found that L-dopa irreversibly incorporates into tubulin, alters mitochondrial transport along axons, and reduces the affinity of the molecular motor KIF5B for Dopatubulin-containing microtubules. This disruption in microtubule functioning impairs mitochondrial traffic, which may have implications for Parkinson's disease patients receiving chronic L-dopa treatment.

Cellular Control of Protein Turnover via the Modification of the Amino Terminus

Nikola Winter et al.

Summary: The first amino acid of a protein plays a significant role in its metabolic stability, with N-degrons mediating turnover. Despite only a few proteases known to contribute to the generation of N-degrons, there is potential for further discovery in many processing paths.
