Interplay between MTOR and GPX4 signaling modulates autophagy-dependent ferroptotic cancer cell death

Yang Liu et al.

Summary: The interplay between the signals of MTOR and GPX4 modulates autophagy-dependent ferroptosis in human pancreatic cancer cells, with GPX4 playing an essential role in inhibiting this process. Depletion of GPX4 enhances the anticancer activity of rapamycin and RSL3 in vitro or in vivo. These findings may lead to the development of new antitumor therapy targeting autophagy-dependent cell death.

Identification of a small molecule as inducer of ferroptosis and apoptosis through ubiquitination of GPX4 in triple negative breast cancer cells

Yahui Ding et al.

Summary: This study identified a novel approach to treat TNBC by inducing ferroptosis through ubiquitination of GPX4 with the compound DMOCPTL, and revealed the role of GPX4 in regulating mitochondria-mediated apoptosis through up-regulation of EGR1 in TNBC cells. Furthermore, the prodrug Compound 13 showed promising anti-tumor effects in vivo with no apparent toxicity, suggesting its potential as a lead compound for developing effective anti-TNBC drugs.

DPP4 gene silencing inhibits proliferation and epithelial-mesenchymal transition of papillary thyroid carcinoma cells through suppression of the MAPK pathway

X. Hu et al.

Summary: The study revealed high expression of DPP4 in PTC tissues, and silencing of DPP4 inhibited PTC cell proliferation and epithelial-to-mesenchymal transition while promoting cell apoptosis through suppression of the MAPK pathway.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Druggable genome and precision medicine in cancer: current challenges

Camille Amandine Dupont et al.

Summary: Recent advances in cancer therapy have provided hope for patients through the development of specific treatment options, but drug resistance remains a challenge. The integration of multidisciplinary approaches has led to the development of next generation therapeutics, and the combination of immune checkpoint inhibitors in synergistic therapeutic regimes offers new promise for cancer patients.

Intratumoral CXCL13+CD8+T cell infiltration determines poor clinical outcomes and immunoevasive contexture in patients with clear cell renal cell carcinoma

Siyuan Dai et al.

Summary: This study revealed the unfavorable clinical outcomes of CXCL13(+)CD8(+)T cells in patients with ccRCC and their impairment of the immune function of total CD8(+)T cells. In addition, CXCL13(+)CD8(+)T cells also showed increased exhausted markers, decreased effector molecules, and better proliferation ability, associated with an immunoevasive tumor microenvironment.

Holo-lactoferrin: the link between ferroptosis and radiotherapy in triple-negative breast cancer

Zheng Zhang et al.

Summary: The study showed that iron-saturated Lactoferrin (Holo-Lf) can induce ferroptosis in triple-negative breast cancer cells and enhance their sensitivity to radiotherapy. Conversely, low iron-saturated Lactoferrin (Apo-Lf) may inhibit this cell death process. Non-triple-negative breast cancer cells may have resistance to Holo-Lf-induced ferroptosis due to higher redox balance capacity.
