Mechanical Feed-Forward Loops Contribute to Idiopathic Pulmonary Fibrosis

Margaret A. T. Freeberg et al.

Summary: Idiopathic pulmonary fibrosis is a progressive disease characterized by accumulation of extracellular matrix in lung tissue, leading to altered mechanical properties. Recent studies suggest that these changes create a feed-forward loop, contributing to fibroblast differentiation and activation. Mechanotransduction pathways, including mechanosensors like integrins and downstream transcription factors, play a role in fibrosis progression.

The serum response factor/cofactors interactions and their implications in diseases

Summary: SRF, a member of the MADS box transcription factor family, is widely expressed in all cell types and regulates downstream genes by interacting with various cofactors. Recent studies on the molecular mechanisms of SRF activation have highlighted its importance in physiological and pathological conditions. This knowledge may provide new opportunities for understanding disease pathogenesis and identifying potential therapeutic targets.

Crosstalk between YAP and TGFβ regulates SERPINE1 expression in mesenchymal lung cancer cells

Hyeon-Joon Kong et al.

Summary: This study found that in mesenchymal lung cancer cells, the expression of SERPINE1 is regulated by the YAP pathway, with TGF-beta treatment enhancing TGF-beta-mediated SERPINE1 expression. The crosstalk between the TGF-beta and YAP pathways underlies SERPINE1 expression in mesenchymal cancer cells.

Serum Response Factor (SRF) Drives the Transcriptional Upregulation of the MDM4 Oncogene in HCC

Rossella Pellegrino et al.

Summary: The transcription factors SRF, ELK1, and ELK4 were identified as drivers of MDM4 expression in hepatocellular carcinoma (HCC). Inhibiting MDM4 transcription with the drug XI-011 reduced the expression of these TFs and MDM4, impacting tumor growth. This study suggests that targeting MDM4 transcription could be a promising approach for future therapy of HCC.

PAI-1 induction during kidney injury promotes fibrotic epithelial dysfunction via deregulation of klotho, p53, and TGF-β1-receptor signaling

Summary: PAI-1 plays a crucial role in tubular dysfunction, leading to fibrosis and cell death through multiple signaling pathways, and is closely associated with klotho, p53, and the TGF-beta RI/SMAD3 axis.
