Development and validation of a four-lipid metabolism gene signature for diagnosis of pancreatic cancer

Yanrong Ye et al.

Summary: This study identified prognostic genes related to lipid metabolism in pancreatic cancer and developed a risk score model with four genes that significantly impact overall survival. The model showed good predictive performance and clinical independence, and was associated with multiple cancer-related pathways. Compared to existing models, this four-gene signature has a higher C-index and greater risk value, making it an effective prognostic indicator for pancreatic cancer survival.

Lipid Metabolism in Oncology: Why It Matters, How to Research, and How to Treat

Yuki Matsushita et al.

Summary: Metabolic reprogramming is a key feature of cancer, where changes in lipid metabolism are now recognized to play an important role in cancer biology. Recent advancements in lipidomics technologies have provided insights into the roles of lipids in cancer, impacting cellular functions and contributing to carcinogenesis. The interaction between cancer cells and the surrounding microenvironment through altered lipid metabolism supports tumor growth and progression, highlighting the potential for novel metabolic targets in cancer treatment.

Loss of G-Protein Pathway Suppressor 2 Promotes Tumor Growth Through Activation of AKT Signaling

Stefanie Chan et al.

Summary: GPS2 functions as a tumor suppressor in breast cancer, and its loss enhances cancer cell proliferation and tumor growth through uncontrolled activation of AKT signaling. The study suggests that GPS2 could serve as a potential biomarker for a subset of breast cancers responsive to PI3K inhibitors.

Prognostic value of lipid metabolism-related genes in head and neck squamous cell carcinoma

Ying Xiong et al.

Summary: In this study, a total of 136 differentially expressed lipid metabolism genes were identified in head and neck squamous cell carcinoma (HNSCC), with 23 of them related to prognosis. Additionally, 11 lipid metabolism-related genes were found to affect clinical features of HNSCC, such as stage, gender, and pathological stage. The prognostic index of lipid metabolism-related genes reflected the state of the HNSCC tumor immune microenvironment.

Lipid signalling enforces functional specialization of Treg cells in tumours

Seon Ah Lim et al.

Summary: Regulatory T cells play a crucial role in immune tolerance and immunosuppression in the tumor microenvironment. Inhibiting lipid synthesis dependent on SREBPs in T-reg cells can enhance antitumor immune responses without autoimmune toxicity, and deletion of SCAP in these cells inhibits tumor growth and enhances immunotherapy. SCAP and SREBPs signaling regulate lipid synthesis and inhibitory receptor signaling in T-reg cells, pointing to new ways of targeting these cells for cancer therapy.

The Prognostic Significance of Anisomycin-Activated Phospho-c-Jun NH2-Terminal Kinase (p-JNK) in Predicting Breast Cancer Patients' Survival Time

Li Chen et al.

Summary: The study found that the expression of p-JNK in breast cancer patients is related to prognosis, with higher levels associated with significantly longer survival times. p-JNK expression is an important prognostic factor that can effectively predict survival in breast cancer patients undergoing NACT. Additionally, treatment with the JNK agonist anisomycin can induce apoptosis and lead to changes in p-JNK and p-STAT3 expression levels.

Adaptive Mechanisms of Tumor Therapy Resistance Driven by Tumor Microenvironment

Peijie Wu et al.

Summary: Cancer is a disease with poor prognosis, and resistance to treatments often hinders clinical outcomes. Tumor microenvironment (TME) plays a crucial role in tumorigenesis, progression, and resistance. Understanding the adaptive mechanisms driven by TME is important for developing effective therapeutic strategies against tumor resistance.

Reprogramming lipid metabolism prevents effector T cell senescence and enhances tumor immunotherapy

Xia Liu et al.

Summary: The functional state of T cells is critical for effective antitumor immunity. T cell senescence driven by malignant tumor cells and regulatory T cells is a common feature in cancers, characterized by unbalanced lipid metabolism and accumulation of lipid droplets. Inhibition of group IVA phospholipase A(2) can reprogram effector T cell lipid metabolism, prevent T cell senescence, and enhance antitumor immunity and immunotherapy efficacy.

The Peripheral Immune Landscape of Breast Cancer: Clinical Findings and In Vitro Models for Biomarker Discovery

Sofia Batalha et al.

Summary: Breast cancer is the deadliest female malignancy worldwide, and defining and analyzing circulating immune profiles of patients is crucial for faster, cheaper, and more precise monitoring of their status. This review compiles knowledge on the peripheral immune profile of breast cancer patients and proposes using this data as a starting point for validating clinically relevant biomarkers of disease progression and therapy response. Further scientific advancement in peripheral biomarker discovery and cancer-immune crosstalk in breast cancer will be instrumental in uncovering molecular mechanisms and potential biomarkers and drug targets.

Functional Assessment of Four Novel Immune-Related Biomarkers in the Pathogenesis of Clear Cell Renal Cell Carcinoma

Daojun Lv et al.

Summary: This study identified immune-related biomarkers IF116, LMNB1, RHBDF2, and TACC3 in ccRCC using bioinformatics analysis, showing their upregulation in the disease and association with tumor stage and grade. These hub genes had great diagnostic and prognostic values for ccRCC, involved in immune signaling pathways, and linked to tumor immune infiltration and checkpoint expressions. The findings were validated by quantitative PCR and immunohistochemical staining.
