Development of Sentinel LN Imaging with a Combination of HAase Based on a Comprehensive Analysis of the Intra-lymphatic Kinetics of LPs

Masaki Gomi et al.

Summary: The study identified that medium-sized anionic LPs accumulate the highest in primary lymph nodes and are captured by CD169-positive macrophages. Fluorescent imaging of sentinel lymph nodes using anionic LPs in a breast cancer model showed high sensitivity. Additionally, co-injection of hyaluronidase significantly improved the detection sensitivity of fluorescent LPs in sentinel lymph nodes.

Characterization of Lipid Nanoparticles Containing Ionizable Cationic Lipids Using Design-of-Experiments Approach

Takeshi Terada et al.

Summary: This study investigated the impact of preparation parameters on the physicochemical properties of LNP-siRNA systems, finding that lipid concentration and flow rate ratio significantly affected particle size and PDI, while dialysis resulted in a decrease in PDI and an increase in particle size.

Graft-derived extracellular vesicles transported across subcapsular sinus macrophages elicit B cell alloimmunity after transplantation

Furong Zeng et al.

Summary: It has been found that donor MHC antigens are released from skin or cardiac allografts via extracellular vesicles (EVs) and captured by macrophages in lymph nodes or spleen, triggering B cell activation and DSA production. This unexpected role of graft-derived EVs opens up possibilities to interfere with EV biogenesis, trafficking, or function to restrict alloreactive B cell priming.

Cationic lipid nanoparticle production by microfluidization for siRNA delivery

Xiaojing Liu et al.

Summary: Microfluidization is a promising method for producing cationic lipid nanoparticles for delivering siRNAs. Optimized characteristics in size reduction and polydispersity can be achieved through experimental design. Positive charge ratio and stability are key factors in determining the structure and efficacy of siRNA lipoplexes.
