Oral preparations with tunable dissolution behavior based on selective laser sintering technique

Yan Yang et al.

Summary: Selective laser sintering (SLS) is used to prepare 3D printlets for tissue engineering and personalized implants. The printability of oral solid preparations was evaluated using a low energy laser, finding that yellow drugs and tartrazine lake can enhance printability. Increasing laser energy density improves printing efficiency but reduces accuracy, with different drugs having different printing mechanisms.

Fused Deposition Modeling (FDM), the new asset for the production of tailored medicines

Sylvain Cailleaux et al.

Summary: The shift towards precision medicine in conventional medicine has led to a focus on customizing oral drug delivery using 3D printing technology, particularly Fused Deposition Modeling (FDM). Various FDM methods have been reported in the production of pharmaceutical solid oral forms, with challenges such as high printing temperatures for thermosensitive ingredients and moisture sensitivity in storage studies. FDM appears to be well-suited for treating rare diseases and populations requiring tailored doses or release kinetics.

Selective Laser Sintering 3-Dimensional Printing as a Single Step Process to Prepare Amorphous Solid Dispersion Dosage Forms for Improved Solubility and Dissolution Rate

Daniel A. Davis et al.

Summary: This study focused on the development of ritonavir-copovidone amorphous solid dispersions (ASDs) using selective laser sintering 3-dimensional (3-D) printing, demonstrating full conversion of the crystalline drug to its amorphous form on a layer-by-layer basis. Key parameters such as powder flow properties, surface temperature, chamber temperature, laser speed, and hatch spacing were crucial for successful ASD formation. The SLS 3-D printed tablets showed a significant increase in solubility, indicating the potential of this technique for creating ASD-based pharmaceutical dosage forms with solubility advantage.

Advances in powder bed fusion 3D printing in drug delivery and healthcare

Atheer Awad et al.

Summary: Powder bed fusion (PBF) is a 3D printing method that selectively consolidates powders into 3D objects using a power source. PBF technologies offer benefits such as superior printing resolution and speed, as well as the ability to create complex products without the need for secondary supports, making them well suited for bespoke drug-laden formulations, devices, and implants.
