Drug Release and Nanodroplet Formation from Amorphous Solid Dispersions: Insight into the Roles of Drug Physicochemical Properties and Polymer Selection

Summary: This study investigated the impact of polymer selection and API physicochemical properties on nanodroplet release from ASDs, finding that HPMCAS resulted in higher drug release compared to PVPVA and low T-g compounds achieved higher levels of nanodroplet formation. Using dynamic light scattering, it was observed that HPMCAS was more effective at stabilizing nanodroplets against size increase. Insights from this study may guide formulation design and excipient selection based on API physicochemical properties.
