Targeting 5-HT2B Receptor Signaling Prevents Border Zone Expansion and Improves Microstructural Remodeling After Myocardial Infarction

J. Caleb Snider et al.

Summary: Inhibiting 5-HT2B can reduce fibrotic scar formation after MI, thereby protecting cardiac structure and function. The redistribution of collagen fibers to thinner, more anisotropic fibers enhances left ventricular contractility, while decreasing fibrotic tissue stiffness limits the hypertrophic response of uninjured cardiomyocytes. Ablation of 5-HT2B in cardiac fibroblasts or myofibroblasts contributes to excessive scar formation and impairs cardiac function after MI.

GRK5 is a regulator of fibroblast activation and cardiac fibrosis

Akito Eguchi et al.

Summary: Pathological remodeling of the heart in chronic heart failure is linked to structural changes perpetuated by cardiac fibroblasts, with GRK5 playing a key role in regulating fibroblast activation and cardiac fibrosis. GRK5 inhibition shows potential beneficial effects in cardiac disease by preventing fibroblast activation and reducing fibrosis and hypertrophy.

AMPKα1 deletion in myofibroblasts exacerbates post-myocardial infarction fibrosis by a connexin 43 mechanism

Cecile Dufeys et al.

Summary: Deleting AMPKα1 in myofibroblasts exacerbates adverse LV remodeling post-MI, enhances fibrotic response, and affects Cx43 expression, potentially through transcriptional and post-transcriptional regulation involving miR-125b-5p.

The double face of miR-320: cardiomyocytes-derived miR-320 deteriorated while fibroblasts-derived miR-320 protected against heart failure induced by transverse aortic constriction

Xudong Zhang et al.

Summary: This study revealed that miR-320 functions differently in various cell types of the heart during the progression of heart failure, with overexpression in CMs exacerbating cardiac dysfunction, and in CFs alleviating cardiac fibrosis and hypertrophy. The effects of miR-320 were mediated through targeting different downstream signaling pathways in CMs and CFs, and could be influenced by upstream regulators such as Ago2 and cell-type-specific transcription factors.
