Contributive Role of TNF-α to L-DOPA-Induced Dyskinesia in a Unilateral 6-OHDA Lesion Model of Parkinson's Disease

Summary: The study aimed to characterize mechanisms regulating neuroinflammation in L-DOPA-induced dyskinesia in mice, finding that astrocytic and microglial responses, as well as increased pro-inflammatory cytokine levels, were associated with LID development. Treatment with cannabidiol and capsazepine reduced LID severity and TNF-alpha levels, suggesting a potential therapeutic approach. Glutamate-induced TNF-alpha release from astrocytes may contribute to LID by modulating corticostriatal glutamatergic inputs, with cannabidiol + capsazepine attenuating this effect.

Doxycycline and its derivative, COL-3, decrease dyskinesia induced by l-DOPA in hemiparkinsonian rats

Summary: The study demonstrates that doxycycline and its analogue compound COL-3 have the potential to reduce l-DOPA-induced dyskinesia in hemiparkinsonian rats. Doxycycline can also suppress the onset of dyskinesia when administered together with l-DOPA, while reducing immunoreactivity and activity of certain proteins in the basal ganglia.
