Differential expression of circular RNAs in bone marrow-derived exosomes from essential thrombocythemia patients

Qiang Wang et al.

Summary: This study identified significant alterations in circRNA profiles in bone marrow-derived exosomes from ET patients, including downregulation of circDAP3, circASXL1, and circRUNX1. Moreover, circRNA-encoding genes and miRNA-mRNA networks targeted by these circRNAs were associated with various biological processes and signaling pathways, suggesting their potential role in ET pathogenesis. Additionally, circ_0014614 was found to inhibit the differentiation of K562 cells into megakaryocytes, indicating a possible mechanism for circRNA-based ET diagnosis and treatment.

Clinical relevance of extracellular vesicles in hematological neoplasms: from liquid biopsy to cell biopsy

Stefania Trino et al.

Summary: Liquid biopsy is becoming increasingly important in oncology within the era of precision medicine, allowing for the isolation and analysis of tumor-derived biomarkers such as extracellular vesicles (EVs) in body fluids. EVs, as lipid bilayer-enclosed particles, have emerged as valuable carriers of cancer information, offering potential as novel biomarkers due to their molecular cargo mirroring parental cell characteristics. The translation of these new diagnostic tools into clinical practice has the potential to deeply revolutionize the field of cancer research.

Distinct profile of CD34+ cells and plasma-derived extracellular vesicles from triple-negative patients with Myelofibrosis reveals potential markers of aggressive disease

Dorian Forte et al.

Summary: In this study, it was found that the absolute numbers of circulating CD34(+) cells were significantly increased in triple-negative (TN) myelofibrosis patients. These cells displayed in vitro defective functions and were unresponsive to the inflammatory microenvironment. Furthermore, the circulating extracellular vesicles (EVs) from TN patients were found to promote the survival of TN CD34(+) cells, suggesting a distinct EV-driven signal promoting malignant hemopoiesis in TN patients. The distinct gene expression profiling analysis and microRNA signature in TN patients may provide insights for novel therapeutic approaches for this rare form of myelofibrosis.

A Broad Overview of Signaling in Ph-Negative Classic Myeloproliferative Neoplasms

Ana Guijarro-Hernandez et al.

Summary: There is growing evidence that Ph-negative myeloproliferative neoplasms are disorders in which multiple signaling pathways are significantly disturbed. The discovery of somatic mutations in JAK2, MPL, and CALR has shed light on the aberrant functions of their mutant products, while additional somatic mutations in other genes have added complexity to the molecular pathogenesis of MPNs. This review highlights the involvement of a wide variety of cellular processes and pathways in the pathogenesis of MPNs, explaining their phenotypic heterogeneity.

Summary: Thrombosis is a major cause of morbidity and mortality in patients with polycythemia vera and essential thrombocythemia. Current understanding of the risk factors and pathophysiology of thrombosis in these patients is growing, though few therapies have shown effectiveness in reducing thrombotic burden. More studies are needed to evaluate the impact of therapeutic agents on thrombotic events in these myeloproliferative neoplasms.

Patients with triple-negative, JAK2V617F- and CALR-mutated essential thrombocythemia share a unique gene expression signature

Samah Alimam et al.

Summary: The study identified similar patterns of gene expression and DNA methylation in patients with essential thrombocythemia (ET) with known driver mutations and those with triple-negative disease, suggesting that the ET phenotype may be primarily driven by transcriptional misregulation and may propagate downstream via MAPK, tumor necrosis factor, and NF kappa B pathways. These findings highlight potential novel mechanisms of disease initiation that require further evaluation.
