Summary: The crosstalk between Notch and MAPK pathway is involved in MEK inhibitor resistance in metastatic melanoma and promotes migration in uveal melanoma cells. The combination of MEK and Notch inhibitors shows synergistic effects in some cells and antagonistic effects in others. In uveal melanoma cells, the combination therapy can block cell migration towards liver cancer cells and induce cells into a senescent-like state.

Summary: This study demonstrates the crucial role of uPAR in the pro-angiogenic activity of melanoma Exos, with uPAR knockout reducing the angiogenic effects of Exos in vitro and in vivo. Inhibition of signaling pathways in endothelial cells, uPAR-EGFR interaction, and EGFR phosphorylation can all contribute to the reduction of melanoma Exos-induced angiogenesis. The identification of uPAR-expressing Exos as a potential biomarker for evaluating angiogenic propensity and treatment response in metastatic melanoma patients is suggested.
