Angiographic investigation of orbital vascular variations in the rabbit and implications for endovascular intra-arterial chemotherapy models

Michael T. Froehler et al.

Summary: The study introduces a refined technique for ophthalmic artery catheterization in New Zealand White rabbits and explores angioanatomical variations in the arterial blood supply to the eyes, with the external ophthalmic artery being dominant in the majority of cases. This provides valuable information for successful replication of studies involving intra-arterial chemotherapy for retinoblastoma treatment in NZWR.

Intravitreal melphalan hydrochloride vs propylene glycol-free melphalan for retinoblastoma vitreous seeds: Efficacy, toxicity and stability in rabbits models and patients

Carley M. Bogan et al.

Summary: The use of intravitreal chemotherapy has significantly improved the treatment of advanced intraocular retinoblastoma. A new propylene glycol-free melphalan formulation shows greater stability, potentially enhancing treatment efficacy and reducing toxicity.

Orbital Swelling in a Child With Retinoblastoma Following Intra-arterial Chemotherapy

Erica L. Friedman et al.

Summary: A 9-month-old girl presented with significant left periocular edema and erythema 3 days after intra-arterial chemotherapy. CT scan showed soft tissue swelling, edema, and retinal detachment. Further evaluation and diagnosis are needed to determine the appropriate intervention.

Effect of Intravenous Chemotherapy Regimen on Globe Salvage Success Rates for Retinoblastoma Based on Disease Class-A Meta-Analysis

Anthony B. Daniels et al.

Summary: Intravenous chemoreduction has been the standard treatment for retinoblastoma, with a shift towards eye salvage and vision preservation goals. Combining IVCRD with intravitreal chemotherapy or switching to intra-arterial chemotherapy is becoming more common, but randomized controlled trials comparing these treatments are lacking. It is essential to establish baseline IVCRD success rates for comparison with new therapies.

Complete preclinical platform for intravitreal chemotherapy drug discovery for retinoblastoma: Assessment of pharmacokinetics, toxicity and efficacy using a rabbit model

Anthony B. Daniels et al.

Summary: Effective melphalan-based intravitreal chemotherapy for retinoblastoma vitreous seeds can cause significant ocular toxicity. This study presents protocols for each step of the drug discovery pipeline to develop novel drugs with maximal efficacy and minimal toxicity. The protocols involve determination of vitreous pharmacokinetics, cytotoxicity assessment against retinoblastoma, calculation of optimal injection dose, determination of maximum-tolerable dose, and evaluation of drug efficacy using a rabbit orthotopic xenograft model.

Intra-arterial Chemotherapy as Primary Treatment for Cavitary Retinoblastoma: Excellent Response in Eight Tumors

Yuxi Zheng et al.

Summary: The study aimed to investigate the treatment response of cavitary retinoblastoma tumors to intra-arterial chemotherapy (IAC). Results showed that all cavitary tumors regressed after IAC treatment, with no recurrence, and 100% globe salvage. Up-front triple therapy and monitoring early signs of regression can help minimize unnecessary additional treatment cycles.
