Summary: In this study, we found abnormal upregulation of PMP22 in TGF-β-activated HSCs and CCl4-induced hepatic fibrosis model in mice, as well as its pro-fibrotic role in aggravating TGF-β-induced HSC activation. Additionally, miR-139-5p targets the 3'UTR of PMP22 and inhibits its expression, thereby hindering TGF-β-induced HSC activation.

PPAR ligands have been explored as potential therapeutic agents for reversing steatohepatitis and liver fibrosis.

Summary: Saroglitazar showed significant efficacy in patients with NAFLD/NASH, improving ALT, liver fat content and insulin resistance. The 4mg dose of Saroglitazar also demonstrated significant improvements in lipid metabolism and lipoprotein particle composition. Overall, Saroglitazar was well-tolerated with a slight weight gain observed in patients.
