Adaptive optics enables aberration-free single-objective remote focusing for two-photon fluorescence microscopy

Yuhan Yang et al.

Summary: Two-photon fluorescence microscopy is widely used for three-dimensional imaging of complex samples. Remote focusing by controlling the divergence of excitation light is a common method, but microscope objectives can introduce aberrations to non-collimated excitation beams, affecting imaging quality. This study used adaptive optics to correct divergence-induced-aberrations and achieve diffraction-limited focal quality for up to 800 μm axial range, demonstrating aberration-free remote focusing for in vivo imaging of neurites and synapses in the mouse brain.

High-definition imaging using line-illumination modulation microscopy

Qiuyuan Zhong et al.

Summary: The development of LiMo technique and HD-fMOST method allows for high-throughput imaging of thick tissues and substantial improvement in neuronal morphology reconstruction. By achieving >30-fold lossless data compression at a voxel resolution of 0.32 x 0.32 x 1.00 mu m(3), real-time high-precision (95% accuracy) brain-wide 3D cell counting is made possible.

Fast, multiplane line-scan confocal microscopy using axially distributed slits

Jean-Marc Tsang et al.

Summary: This study presents a multiplane line-scan imaging strategy using reflecting slits to probe different depths, enabling simultaneous imaging of optically sectioned image stack with a single camera at frame rates of hundreds of hertz. The system is demonstrated to be applicable for monitoring fast dynamics in biological samples.
