Targeting Lysyl Oxidase Family Meditated Matrix Cross-Linking as an Anti-Stromal Therapy in Solid Tumours

Yordanos F. Setargew et al.

Summary: The lysyl oxidase (LOX) family of enzymes play a key role in the formation of desmoplastic matrix in both primary tumors and secondary metastatic sites. With the growing interest in anti-stromal therapies, the LOX family has emerged as a potential clinical target. This review explores how dysregulation of the lysyl oxidase family in solid cancers contributes to disease progression, as well as the potential use of the LOX family as diagnostic and prognostic markers in solid tumors.

A collagen glucosyltransferase drives lung adenocarcinoma progression in mice

Hou-Fu Guo et al.

Summary: Guo et al. have determined the molecular basis of collagen lysyl hydroxylase 2 (LH2) substrate specificity, showing that LH2 functions not only as a telopeptidyl lysyl hydroxylase, but also as a collagen glucosyltransferase to drive lung cancer progression. The research suggests that LH2 plays a crucial role in promoting LUAD progression through a variety of mechanisms, including tLH-dependent and tLH-independent pathways.

Abnormal Bone Collagen Cross-Linking in Osteogenesis Imperfecta/Bruck Syndrome Caused by Compound Heterozygous PLOD2 Mutations

Charlotte Gistelinck et al.

Summary: Bruck syndrome is a congenital disorder clinically overlapping with osteogenesis imperfecta, caused by biallelic mutations in FKBP10 or PLOD2. Research on a 4-year-old boy with BS caused by compound heterozygous mutations in PLOD2 found abnormal collagen cross-linking in bone, shedding light on the complex mechanisms behind brittle bones caused by defective collagen cross-linking.
