Repression of the stress granule protein G3BP2 inhibits immune checkpoint molecule PD-L1

Summary: This study reveals the role of stress granule-associated protein G3BP2 in regulating the immune checkpoint molecule PD-L1 in breast and glioblastoma cancer cells, suggesting a connection between stress granule formation and immune checkpoint control in cancer. Inhibition of G3BP2 leads to decreased PD-L1 expression, enhancing the immune response and improving the survival of breast tumor-bearing mice.

Deficiency in G3BP1 can lead to phenotypes related to mTORC1 hyperactivity, and low G3BP1 levels in tumors enhance mTORC1-driven cell motility and correlate with poor outcomes in patients. Additionally, inhibition of G3bp1 in zebrafish affects neuronal development and function.
