Immune and TRG repertoire signature of the thymus in Down syndrome patients

Shira Rabinowicz et al.

Summary: Patients with Down syndrome have an increased diversity in T cell receptor gamma repertoire but decreased clonal expansion compared to non-syndromic patients. The V and J gene usages are similar between the two groups, indicating that differences in TRG repertoire may contribute to DS patients' predisposition to infections and autoimmune diseases. Using next-generation sequencing techniques can reveal repertoire abnormalities not seen with previous methods.

Rubina Mulchandani et al.

Summary: Studies have shown significantly elevated levels of pro-inflammatory cytokines and reduced T lymphocytes in severe COVID-19 cases. However, there is heterogeneity across studies due to small sample sizes, nonuniform outcome assessment, and varied definitions of disease severity, indicating a need for well-designed and adequately powered prospective studies for more definitive answers.

Individuals with Down syndrome hospitalized with COVID-19 have more severe disease

Louise Malle et al.

Summary: This study analyzed individuals with Down syndrome who were hospitalized with COVID-19 in New York City and found that they were on average ten years younger than non-DS patients and had more severe disease, particularly with an increased incidence of sepsis and mechanical ventilation. Special care should be considered for both the prevention and treatment of COVID-19 in these patients.

Baricitinib plus Remdesivir for Hospitalized Adults with Covid-19

A. C. Kalil et al.

Summary: Combination treatment with baricitinib plus remdesivir was superior to remdesivir alone in reducing recovery time and accelerating improvement in clinical status among patients with Covid-19, notably among those receiving high-flow oxygen or noninvasive ventilation. The combination was associated with fewer serious adverse events.

Summary: Baricitinib is believed to have a dual effect on SARS-CoV2 infection by reducing inflammatory response and receptor-mediated viral endocytosis. This study aims to evaluate the effectiveness and safety of using baricitinib to prevent respiratory insufficiency progression in patients with SARS-CoV2 pneumonia.

Summary: Health conditions, immune dysfunction, and premature aging associated with trisomy 21 (Down syndrome, DS) may impact the clinical course of COVID-19. The study found that individuals with DS have similar signs/symptoms and risk factors for severe disease course as the general population, but higher rates of medical complications and mortality, especially from age 40.
