Integrative Single-Cell RNA-Seq and ATAC-Seq Analysis of Human Developmental Hematopoiesis

Summary: Using single-cell RNA sequencing and single-cell assay for transposase-accessible chromatin sequencing, researchers identified three highly proliferative oligopotent progenitor populations downstream of hematopoietic stem cells (HSCs)/multipotent progenitors (MPPs) in human fetal liver and bone marrow. They observed dynamic changes in chromatin accessibility and lineage-specific transcription factor activity along the differentiation trajectory, providing a useful framework for future investigation of human developmental hematopoiesis.

Summary: In adult mammals, hematopoiesis is tightly regulated by signals from the microenvironment called niche. The vascular system acts as a second niche to control lymph gland homeostasis by regulating intracellular calcium levels through the FGF signaling pathway. This study shows that two niches contribute to the control of Drosophila blood cell homeostasis through differential regulation of progenitors.
