Summary: The study found that microsatellites tend to shorten over time, while MMRd introduces novel mutations throughout the disease course, contributing to heterogeneity. Some microsatellites are altered among all subclones in a patient, whereas other loci are only altered in particular subclones corresponding to subclonal phylogenetic relationships. Overall, the results suggest that MMRd is a major driver of heterogeneity, leading to both MSI and subclonal divergence.

Genomic and prognostic heterogeneity among RAS/BRAFV600E/TP53 co-mutated resectable colorectal liver metastases

Summary: This study investigated the potential for improved prognostic stratification for patients with colorectal liver metastases undergoing hepatic resection by combined biomarker analysis with DNA copy number aberrations (CNAs). The results showed that RAS/BRAF(V600E) and TP53 co-mutations were associated with a poor patient outcome, and CNAs also had prognostic associations. Combined analysis of RAS/BRAF(V600E)/TP53 mutations and CNAs identified patients with particularly poor outcomes.

Summary: This study revealed the significant role of copy number aberrations in cancer evolution, as well as the importance of neoantigens and diverse T cell receptor repertoire in postoperative recurrence. The comparison of driver mutations and immune response-related molecules between early and metastatic sites showed differences, and the postoperative recurrence was associated with neoantigen levels and TCR diversity in the metastatic sites.

Breast tumours maintain a reservoir of subclonal diversity during expansion

Summary: Breast tumors and cell lines consist of numerous subclones organized into a few major superclones. Following clonal TP53 mutations, loss-of-heterozygosity events, and genome doubling, primary tumors undergo a period of transient genomic instability, with ongoing copy number evolution during expansion. Subcloned daughter cells demonstrate genomic rediversification, showing that triple-negative breast cancers continually evolve chromosome aberrations and maintain subclonal diversity during primary tumor growth.

Tracing oncogene-driven remodelling of the intestinal stem cell niche

Summary: The study utilized a multicolour reporter mouse model, the Red2Onco system, to investigate interactions between tumour cells and the microenvironment. The research revealed that oncogene-driven paracrine remodeling leads to the destruction of wild-type tissue and promotes region transformation dominated by oncogenic clones. Mechanisms such as secretion of BMP ligands and alteration of the WNT signaling environment contribute to this process.

NOTUM from Apc-mutant cells biases clonal competition to initiate cancer

Summary: The tumour suppressor APC is the most commonly mutated gene in colorectal cancer. Loss of Apc in intestinal stem cells drives the formation of adenomas in mice via increased WNT signalling, while reduced secretion of WNT ligands increases the ability of Apc-mutant intestinal stem cells to colonize a crypt. The study found that Apc-mutant cells are enriched for transcripts encoding several secreted WNT antagonists, with Notum being the most highly expressed. These cells actively inhibit the proliferation of surrounding wild-type crypt cells, outcompeting them in the niche and driving their differentiation.

Apc-mutant cells act as supercompetitors in intestinal tumour initiation

Summary: In the ISC niche, the delicate balance of WNT agonists and antagonists is crucial for maintaining the ISC compartment and preventing unrestrained activation of the WNT pathway. Apc-mutant ISCs function as supercompetitors by secreting WNT antagonists to induce differentiation of neighboring wild-type ISCs. Lithium chloride prevents the expansion of Apc-mutant clones and adenoma formation by rendering wild-type ISCs insensitive to WNT antagonists through downstream activation of WNT by inhibiting GSK3 beta. Boosting the fitness of healthy cells may be a powerful strategy to limit the formation of cancers in high-risk individuals.
