Integrative Single-Cell RNA-Seq and ATAC-Seq Analysis of Human Developmental Hematopoiesis

Anna Maria Ranzoni et al.

Summary: Using single-cell RNA sequencing and single-cell assay for transposase-accessible chromatin sequencing, researchers identified three highly proliferative oligopotent progenitor populations downstream of hematopoietic stem cells (HSCs)/multipotent progenitors (MPPs) in human fetal liver and bone marrow. They observed dynamic changes in chromatin accessibility and lineage-specific transcription factor activity along the differentiation trajectory, providing a useful framework for future investigation of human developmental hematopoiesis.

Erythrocytes 3D genome organization in vertebrates

Anastasia Ryzhkova et al.

Summary: The lack of 3D interaction patterns such as loops or TADs in the blood cells of various vertebrate species was observed, along with large-scale chromatin rearrangements in mammals, birds, reptiles, and amphibians displaying a distinct second diagonal pattern indicating increased long-range contacts unrelated to TADs or compartments. These principles of genome organization were found to be conservative in vertebrate erythroid cells.
