Characterization of hereditary red blood cell membranopathies using combined targeted next-generation sequencing and osmotic gradient ektacytometry

Joan-Lluis Vives-Corrons et al.

Summary: The combination of t-NGS and LoRRca has proven to be effective in identifying mutations in RBC membranopathies in patients with hereditary hemolytic anemia, contributing to a better understanding of the role of RBC rheology in anemia severity. Out of 42 patients studied, 83.8% had mutations identified, while 26.2% remained undiagnosed despite suspected membranopathy.

Rapid Gardos Hereditary Xerocytosis Diagnosis in 8 Families Using Reticulocyte Indices

Veronique Picard et al.

Summary: Gardos channelopathy, a rare hereditary hemolytic anemia, can be diagnosed faster by studying reticulocyte indices, which showed distinct differences in rMCV and rMCHC values compared to normal or Piezo1-HX patients. These parameters may serve as a useful tool for interpreting new KCNN4 variants in diagnosis.

Screening tools for hereditary hemolytic anemia: new concepts and strategies

Elisa Fermo et al.

Summary: Recent advancements in specific molecules and treatments for hereditary hemolytic anemias have increased the focus on differential diagnosis and the need for novel diagnostic assays and devices. Automated blood cell analyzers, omic-approaches including NGS technologies, and the development of new automated tools based on artificial neural networks are considered the future strategies in this field.

Identification of protein quality control regulators using a Drosophila model of TPI deficiency

Stacy L. Hrizo et al.

Summary: Triosephosphate isomerase (TPI) deficiency is a rare metabolic disorder characterized by reduced TPI protein stability and potential co-translational selection for polyubiquitination. A genome-wide RNAi screen targeting quality control proteins identified novel regulators of TPI(sugarkill) degradation, uncovering potential therapeutic pathways for TPI Df and other diseases.
