Identifying Drug Response by Combining Measurements of the Membrane Potential, the Cytosolic Calcium Concentration, and the Extracellular Potential in Microphysiological Systems

Karoline Horgmo Jaeger et al.

Summary: By combining multi electrode arrays (MEAs) with optical measurements, it is possible to accurately estimate the current density and conduction velocity of sodium channels in cardiomyocytes, which advances the research on the application of novel drugs in cardiac cells.

In vitro safety clinical trial of the cardiac liability of drug polytherapy

Berenice Charrez et al.

Summary: A chronic preclinical drug screening platform using a cardiac microphysiological system was utilized to assess the cardiotoxicity of repurposed drugs HCQ and AZM for SARS-CoV-2, showing that polytherapy resulted in increased QT interval but decreased arrhythmias compared to monotherapy, in line with clinical trial outcomes. This system accurately predicted clinical arrhythmias associated with QT prolongation and rhythm instabilities, providing a valuable tool for clinicians to design trials and define monitoring biomarkers for safe COVID-19 therapeutics.

An evaluation of co-use of chloroquine or hydroxychloroquine plus azithromycin on cardiac outcomes: A pharmacoepidemiological study to inform use during the COVID19 pandemic

Scott M. Vouri et al.

Summary: This study compared the risk of cardiac adverse events when using combinations of chloroquine with azithromycin or amoxicillin. The results showed that the combination of routine doses of chloroquine and azithromycin did not significantly increase arrhythmias in a real-world population, but the conclusions were limited due to a small sample size and outcome rates. Further research is needed.
