Structure-based drug repurposing for targeting Nsp9 replicase and spike proteins of severe acute respiratory syndrome coronavirus 2

Vaishali Chandel et al.

Summary: Drug re-purposing using bioinformatics approach identified FDA-approved drugs that can potentially inhibit the Nsp9 replicase and spike proteins in SARS-CoV-2. Molecular dynamics and docking were used to screen and select hit compounds with high binding energy and favorable ADME profile. Conivaptan and Tegobuvir showed promising potential as therapeutic agents against SARS-CoV-2, with further validation needed for their efficacy.

Natural products may interfere with SARS-CoV-2 attachment to the host cell

Abdo A. Elfiky

Summary: The study tested natural product compounds against the HSPA5 substrate-binding domain beta (SBD beta), the recognition site for the SARS-CoV-2 spike, showing high to moderate binding affinity for phytoestrogens and estrogens which may interfere with SARS-CoV-2 attachment to stressed cells, potentially serving as anti-COVID-19 agents for high-risk individuals such as elders, cancer patients, and front-line medical staff.

Discovery of potential multi-target-directed ligands by targeting host-specific SARS-CoV-2 structurally conserved main protease

Rakesh S. Joshi et al.

Summary: The study indicates structural divergence of the main protease (M-Pro) within the Coronaviridae family but with conserved structural features. Researchers identified several natural molecules, such as delta-viniferin, myricitrin, that strongly bind to SARS-CoV-2 M-Pro and other potential targets. This approach of multi-target-directed ligand identification may provide new avenues for drug discovery against SARS-CoV-2 infection.

Withanone and caffeic acid phenethyl ester are predicted to interact with main protease (Mpro) of SARS-CoV-2 and inhibit its activity

Vipul Kumar et al.

Summary: The study showed that Withanone (Wi-N) and Caffeic Acid Phenethyl Ester (CAPE) can bind to the SARS-CoV-2 M(pro) enzyme with similar efficacy and binding energy to a known protease inhibitor. This suggests that these natural compounds may have the potential to inhibit the functional activity of the virus, offering a possible therapeutic value for managing COVID-19. Further validation in laboratory and clinical tests is warranted.

Summary: This study evaluated the anti-viral efficacy of 27 caffeic-acid derivatives against COVID-19 through molecular docking and molecular dynamics simulation. Five natural bioactive entities were identified as potent modulators of COVID-19, showing potential for developing phytonutrients-based antiviral therapeutics. Further studies are recommended to assess the antiviral effects of these phytochemicals against SARS-CoV-2 in in vitro and in vivo models.
