Stiffening transition in asymmetric lipid bilayers: The role of highly ordered domains and the effect of temperature and size

Summary: Cellular membranes are composed of a variety of lipids and proteins with different compositions between the two leaflets, leading to differential stress and affecting mechanical properties. The threshold asymmetry increases with temperature, potentially restricted to a limited range above the gel transition. Stiffening transition is more readily observed in larger membranes with smaller curvature.

Investigation of the domain line tension in asymmetric vesicles prepared via hemifusion

Summary: The plasma membrane is asymmetric in lipid composition, leading to different lipid-lipid interactions and properties in the exoplasmic and cytoplasmic leaflets. The external leaflet can form ordered and disordered fluid domains, while the cytoplasmic leaflet tends to form a single fluid phase. Research on interleaflet interactions shows that the appearance of modulated phases indicates a decrease in domain line tension.

Simulating realistic membrane shapes

Summary: Biological membranes exhibit diversity in their shapes and complexity in chemical compositions that are linked to many cellular functions. Simulation studies using a diverse range of computational techniques are able to capture this complexity at increasing levels of realism and connect the microscopic view on protein-lipid interactions to cellular morphologies at the level of entire organelles. Recent advances highlight the understanding of this complexity, identify current bottlenecks, and sketch possible ways ahead.

Quantitative relationship between cholesterol distribution and ordering of lipids in asymmetric lipid bilayers

Summary: Research shows that the distribution of cholesterol in cell membrane leaflets is solely dependent on the ordering of lipids, with the ratio of cholesterol amount matching the ratio of lipid order in the two leaflets. Transfer of cholesterol between leaflets in asymmetric bilayers can alleviate stresses induced by differences in lipid order.
