Genetic architecture of schizophrenia: a review of major advancements

Sophie E. Legge et al.

Summary: Schizophrenia is a severe psychiatric disorder with high heritability. Recent research has identified numerous genetic loci associated with the disorder, with genome-wide association studies revealing 176 distinct genetic loci and ongoing studies indicating a total of 270 common genetic loci. Polygenic risk scores can explain a portion of the variance in schizophrenia, and rare variant studies have highlighted specific genetic variants linked to an increased risk of schizophrenia. Gene-set analyses have also shown enrichment of genetic variants related to schizophrenia in synaptic pathways, pointing towards potential new avenues for research and treatment.

The genetic basis of major depression

K. M. Kendall et al.

Summary: Major depressive disorder (MDD) is a common, debilitating disorder with heritability ranging from 30% to 50%. Studies have confirmed the highly polygenic nature of MDD, identifying over 100 genetic risk loci. Rare copy number variants associated with MDD risk have also been recently discovered.

Early-life stress effects on BDNF DNA methylation in first-episode psychosis and in rats reared in isolation

Helene A. Fachim et al.

Summary: The study suggests that childhood maltreatment may result in increased BDNF methylation, providing a mechanism underlying the association between early-life stress and psychosis.
