Summary: This study reveals that Vpr antagonizes cellular restriction against HIV-1 by targeting SLF2 and the SMC5/6 complex. Depletion of SMC5/6 or SLF2 increases viral expression by enhancing chromatin accessibility of unintegrated virus, implicating the SMC5/6 complex in nuclear immunosurveillance and providing a conserved antagonism mechanism by Vpr.
