Mu opioid receptor in microglia contributes to morphine analgesic tolerance, hyperalgesia, and withdrawal in mice

David Reiss et al.

Summary: One major challenge in medicine is to develop potent pain therapies without the adverse effects of opiates. This study investigated the role of microglial mu opioid receptor (MOR) in opiate analgesic tolerance, opioid-induced hyperalgesia (OIH), and physical dependence. The researchers found that deleting MOR in microglia delayed morphine analgesic tolerance and abolished OIH in mice. Their results suggest that blocking microglial MOR could be a therapeutic target for opiate analgesia without the opiate adverse effects.

Delta opioid receptor activation modulates affective pain and modality-specific pain hypersensitivity associated with chronic neuropathic pain

Catherine M. Cahill et al.

Summary: Delta opioid receptor agonists alleviate neuropathic pain and have minimal effect on sensory thresholds in the absence of injury. Nerve injury enhances the function and localization of DORs, and endogenous DOR activity plays a role in suppressing pain associated with the injury.

Astrocytes mediate the effect of oxytocin in the central amygdala on neuronal activity and affective states in rodents

Jerome Wahis et al.

Summary: This study reveals a morphologically distinct subpopulation of astrocytes expressing OT receptors in the central amygdala of mice and rats, mediating the anxiolytic and positive reinforcement effects of OT. It challenges the traditional belief that OT exclusively acts on neurons, highlighting the essential role of astrocytes in modulating emotional states under normal and chronic pain conditions.

Considering sex as a biological variable will require a global shift in science culture

Rebecca M. Shansky et al.

Summary: For over half a century, male rodents have been the default model organism in preclinical neuroscience research, leading to higher misdiagnosis and adverse effects in women. Studying both sexes is crucial, but incentives and funding structures often discourage researchers. Calls for cultural and structural changes in research and evaluation processes are needed to create a more equitable knowledge base.

Generation of the Human Pluripotent Stem-Cell-Derived Astrocyte Model with Forebrain Identity

Ulla-Kaisa Peteri et al.

Summary: The study established a method to generate human astrocytes from hPSCs with forebrain patterning and CNTF, confirming activation of forebrain differentiation pathways at D30 and D60 of differentiation in vitro. The reproducibility of the method across multiple hPSC lines and the usefulness of forebrain astrocyte modeling in research investigating forebrain pathology were demonstrated. Intracellular calcium responses to ATP indicated differentiation of the functional astrocyte population with specific gene expressions.

Sexually Dimorphic Immune and Neuroimmune Changes Following Peripheral Nerve Injury in Mice: Novel Insights for Gender Medicine

Valentina Vacca et al.

Summary: This study identified gender differences in immune responses to neuropathic pain, with females showing higher infiltration of T cells in nerves and males showing higher infiltration of macrophages. Additionally, cytokine and chemokine levels were differently regulated in blood and nerve lysates from female mice compared to male mice. These findings provide insights into gender-associated inflammatory profiles in neuropathy and could contribute to the development of differential biomarkers and personalized medicine.
