Phages as immunomodulators and their promising use as anti-inflammatory agents in a cftr loss-of-function zebrafish model

Marco Cafora et al.

Summary: The study demonstrates that the phage cocktail exhibits anti-inflammatory effects in cystic fibrosis animal models, reducing the expression of pro-inflammatory markers. This anti-inflammatory effect is mainly achieved through the activation of the Toll-like Receptor pathway, and the phages significantly reduce neutrophil migration after acute inflammatory induction.

Overcoming Challenges to Make Bacteriophage Therapy Standard Clinical Treatment Practice for Cystic Fibrosis

Renee N. Ng et al.

Summary: This review discusses the use of primary airway epithelial cells to represent the mucoenvironment of the CF lungs for validating the safety and efficacy of phage therapy. The combination of phage therapy and antimicrobials is gaining attention for delaying the onset of MDR infections. Efforts to translate phage therapy into standard clinical practice have gained traction in the past 5 years, calling for collaboration and standardized policies.

The Safety and Toxicity of Phage Therapy: A Review of Animal and Clinical Studies

Dan Liu et al.

Summary: Phage therapy has garnered renewed interest due to increasing rates of antibiotic resistant bacteria infections. Safety and toxicity data associated with phage therapy in animals and humans have been systematically reviewed from English language publications between 2008-2021. While some adverse events were found in relation to phage therapy, serious events were extremely rare, highlighting the need for structured safety and tolerability endpoints in the therapeutic use of phages. Comprehensive and standardized reporting of potential toxicities associated with phage therapy has generally been lacking in published literature.

Summary: Currently, the application of phage therapy in clinical practice is hindered by a lack of standardized protocols, limited information on systemic effects, and previous heterogeneity in phage selection and combination as part of phage cocktails, resulting in insufficient safety and efficacy data for phage therapy.
